The Argentine got off to a great start and won the first two sets without much effort.
Thiem also came back from a set tiebreaker to win the next and tie things up.
The decider also belonged to Pedro Cacin, who sent the former US Open 2020 winner home.
Dominic Thiem also lost the Roland Garros final in 2019.
In the second round, Pedro Cacin will face No. 15 seed Borna Coric of Croatia, who outlasted Federico Coria in four sets 7-6, 6-7, 6-3, 6-3.
The Roland Garros tournament takes place from May 28 to June 11, has a total prize pool of 49.6 million euros, and is the 127th tournament. Iga Sviatek and Rafael Nadal (14-time winner in Paris) became the champions of the title.
