Zverev (10 ATP) could not make it to the second round of the tournament, following the Dane Holger Run, overtaken by the 122nd place in the world.
Safiullin completely dominated both sets and secured qualification after just one hour and 16 minutes of play.
Roman Safiullin will meet American Ben Shelton (20 ATP) in the third round.
The Shanghai Masters will be held from October 4 to 15 in hard mode and offers $8,800,000 in prize money. The champion of the title is Daniil Medvedev.
